چکیده
EGFR inhibitors (EGFRIs) frequently cause cutaneous adverse events such as acneiform rashes. However, the pathogenetic mechanism of remains unclear. We previously reported that EGFRIs suppressed expression human β-defensin-3 (hBD3) in patients treated with and cultured keratinocytes induced by S. epidermidis via Toll-like receptor 2 (TLR2). Because staphylococci are commonly detected skin lesions, we hypothesized commensal maintain innate immune response inducing hBD3 EGF-signaling pathway. To clarify pathogenesis EGFRI-induced eruptions, analyzed whether exposed to produce EGF. First, determine ligands induce hBD3, were administered into keratinocyte culture medium. The well epidermidis. Furthermore, markedly increased TGF-α. and/or TGF-α administrated medium presence or absence EGFRI verify suppressive effects on was stimulated increase completely inhibited EGFRI. Next, is TLR2, TLR2-knockdown epidermidis, and levels measured. significantly keratinocytes. In addition, also inhibition TLR2. Our results suggest under normal condition, but EGFRIs, decreased, resulting eruptions.

Drug-Induced Acneiform Eruptions
Acne vulgaris is a chronic skin disease that develops as a result of inflammation of the pilosebaceous unit and its clinical course is accompanied by comedones, papules, pustules, and nodules. A different group of disease, which is clinically similar to acne vulgaris but with a different etiopathogenesis, is called “acneiform eruptions.” In clinical practice, acneiform eruptions are generally t...

Dear Editors, Epidermal growth factor receptor (EGFR) inhibitors such as erlotinib and gefitinib have been established as effective therapies for non-small cell lung cancer, pancreatic cancer, colorectal cancer, and head and neck cancer (1). However, a number of skin manifestations, such as acneiform papules, xerosis, photosensitive dermatitis, pruritus, perionychia and vasculitis have been rep...

Acneiform rash is the most common side effect of epidermal growth factor receptor (EGFR) inhibitors (EGFRis), and it occurs in 50–100% of patients. This condition can affect the quality of life of these patients and can sometimes lead to a discontinuation of the antineoplastic therapy. Several recent prospective studies have addressed and evaluated different interventions to mitigate or reduce ...
